Tumor fibrosis models in translational research<\/h2>\n \n\n <\/div>\n

Cancer-associated fibroblasts (CAFs)<\/a>\u00a0play a crucial role in creating a pro-tumorigenic microenvironment by altering extracellular matrix (ECM) deposition and remodeling (desmoplasia).<\/p>\n

This dense stroma can enable cancer cells to evade growth suppressors, promote invasion and metastasis, act as a barrier to drug diffusion, and reduce immune responses. As such, targeting CAFs and CAF-derived ECM presents a promising strategy for optimizing cancer therapies.<\/p>\n

However, the heterogeneity of CAFs within and between individual tissues\u2014including their collagen production and contribution to desmoplasia\u2014poses significant challenges to this approach.<\/p>\n <\/div>\n <\/div>\n

The Scar-in-a-Jar (SiaJ) model\u2014a powerful tool for CAF research<\/h2>\n

The Scar-in-a-Jar (SiaJ) model is a straightforward in vitro tool that allows us to study CAF biology and assess the direct impact of therapeutic interventions, especially those related to ECM remodeling. At Nordic Bioscience, we offer a repertoire of CAFs derived from pancreatic, lung, colon, and breast cancer.<\/p>\n

These CAFs can be stimulated with various pro-fibrotic compounds or treated with anti-fibrotic drugs. By collecting supernatant over 12 days and measuring our proprietary\u00a0Nordic ProteinFingerPrint\u2122 biomarkers<\/a>, we gain valuable insights into distinct\u00a0tumor microenvironments.<\/a><\/p>\n <\/div>\n

Testing efficacy of anti-fibrotic compounds in SiaJ<\/h2>\n \n\n <\/div>\n

The SiaJ model is also an effective platform for testing the efficacy of anti-fibrotic compounds. For example, the small molecule ALK5i, a TGF-\u03b2 receptor inhibitor, reduces\u00a0PRO-C3 (nordicPRO-C3\u2122)<\/a>\u00a0levels to baseline or even lower after treatment. Additionally, biological compounds such as the anti-TGF-\u03b2 antibody Fresolimumab demonstrate efficacy in the SiaJ model, by reducing\u00a0PRO-C1,\u00a0nordicPRO-C3\u2122<\/a>\u00a0and\u00a0nordicPRO-C6\u2122<\/a> in a dose-dependent manner, dependent on the CAF type.<\/p>\n
